<p id="p-2">The indexed individual, from now on termed M116, was the world’s oldest verified living person from January 17<sup>th</sup><span> </span>2023 until her passing on August 19<sup>th</sup><span> </span>2024, reaching the age of 117 years and 168 days (<a href="https://www.supercentenarian.com/records.html">https://www.supercentenarian.com/records.html</a>). She was a Caucasian woman born on March 4<sup>th</sup><span> </span>1907 in San Francisco, USA, from Spanish parents and settled in Spain since she was 8. A timeline of her life events and her genealogical tree are shown in<span> </span><strong>Supplementary Fig. 1a-b</strong>. Although centenarians are becoming more common in the demographics of human populations, the so-called supercentenarians (over 110 years old) are still a rarity. In Catalonia, the historic nation where M116 lived, the life-expectancy for women is 86 years, so she exceeded the average by more than 30 years (<a href="https://www.idescat.cat/">https://www.idescat.cat</a>). In a similar manner to premature aging syndromes, such as Hutchinson-Gilford Progeria and Werner syndrome, which can provide relevant clues about the mechanisms of aging, the study of supercentenarians might also shed light on the pathways involved in lifespan. To unfold the biological properties exhibited by such a remarkable human being, we developed a comprehensive multiomics analysis of her genomic, transcriptomic, metabolomic, proteomic, microbiomic and epigenomic landscapes in different tissues, as depicted in<span> </span><strong><a id="xref-fig-1-1" class="xref-fig" href="https://www.biorxiv.org/content/10.1101/2025.02.24.639740v1#F1">Fig. 1a</a></strong>, comparing the results with those observed in non-supercentenarian populations. The picture that emerges from our study shows that extremely advanced age and poor health are not intrinsically linked and that both processes can be distinguished and dissected at the molecular level.</p>

<h2 class="ft-mol-subtitle ft-mol-subtitle--listSimple ft-helper-fontColor-quaternary">Un equipo liderado por el doctor Manel Esteller ha dado con las claves de la longevidad de la 'abuela de Catalunya'</h2>
<p class="ft-text"><strong>Maria Branyas</strong><span> </span>murió el pasado<span> </span><strong>19 de agosto a los 117 años</strong>, marca que la había convertido en la persona<span> </span><strong>más anciana del mundo</strong>. A pesar de su avanzada edad, conservaba la lucidez intacta y apenas sufría dolor articular y sordera.</p>
<p class="ft-text">En búsqueda de las claves de la longevidad, el científico Manel Esteller, jefe del Grupo de Epigenética del Institut Josep Carreras, estudió su genes y ahora cree haber dado con el 'secreto Branyas'. Según ha explicado a TV3, la superanciana había<span> </span><strong>heredado un genoma</strong><span> </span>que hacía que sus<span> </span><strong>células equivalieran a las de una persona de "entre 100 y 102 años"</strong>. Y, además, contaba con una<span> </span><strong>microbiota intestinal</strong><span> </span>con un número de bifidobacterias<strong><span> </span>parecida a la de un niño pequeño.</strong></p>
<p><span>Varios equipos de investigación son los implicados en el análisis del organismo de Branyas, del que se han estudiado desde las </span><strong>mutaciones genéticas</strong><span>, los </span><strong>tejidos<span> </span></strong><span>e, incluso, sus </span><strong>heces<span> </span></strong><span>para esclarecer los factores de su </span><strong>extraordinaria longevidad</strong><span>.</span></p>
<p class="ft-text">El estudio hecho a Maria Branyas, que vivió hasta los 117 años en buen estado de salud, demuestra que<strong><span> </span>la vejez no va necesariamente asociada a la enfermedad</strong>, ya que, según los investigadores, su genoma hacía que sus células "se sintieran" y "se comportaran" como<span> </span><strong>células más jóvenes</strong>, "con una edad biológica de unos<span> </span><strong>diecisiete años menos</strong>".</p>
<p class="ft-text">Según ha indicado Manel Esteller en unas declaraciones a EFE, el<span> </span><strong>genoma privilegiado<span> </span></strong>de Maria Branyas<span> </span><strong>heredado<span> </span></strong>de su padre y de su madre "le concedía una<span> </span><strong>gran protección frente a enfermedades cardiovasculares</strong><span> </span>y de otros tipos, y su microbiota hacía que tuviera una<strong><span> </span>baja inflamación intestinal</strong>".</p>
<h3 class="ft-helper-spacer-t-xs ft-helper-fontSize-heading-XS">Una microbiota propia de una niña</h3>
<p class="ft-text">De hecho, ha precisado Esteller, Maria Branyas "tenía una<span> </span><strong>microbiota intestinal como la de una niña</strong>", por lo que ambos factores, el genoma privilegiado y una microbiota muy sana, hacían que hubiera un<span> </span><strong>decalaje entre su edad cronológica y la biológica</strong>".</p>
<p><span>Manel Esteller, que está considerado como una de las máximas autoridades en epigenética y en el estudio del envejecimiento y el cáncer, considera que el estudio genético a Maria Branyas supone la investigación</span><strong><span> </span>"más exhaustiva y completa"</strong><span> que se ha hecho a una persona </span><strong>supercentenaria</strong><span>. "Hay bastantes personas centenarias en el mundo pero pocas supercentenarias, que son las que</span><strong><span> </span>sobrepasan los 110 años",</strong><span> ha aclarado Esteller.</span></p>
<p class="ft-text">En el estudio, participaron equipos de investigadores de prestigio internacional, y se llevó a cabo porque estaban<span> </span><strong>convencidos del interés</strong><span> </span>que podía tener el análisis de los genes de la abuela supercentenaria.</p>
<p class="ft-text">Unos análisis que, junto a los genéticos, también estudieron<span> </span><strong>otros parámetros<span> </span></strong>de su organismo, como los<span> </span><strong>tejidos<span> </span></strong>y la<span> </span><strong>microbiota intestinal</strong><span> </span>y que, en su conjunto, han aportado<span> </span><strong>datos muy valiosos</strong><span> </span>para los científicos para ayudar a explicar los<strong><span> </span>mecanismos del envejecimiento</strong>.</p>
<h3 class="ft-helper-spacer-t-xs ft-helper-fontSize-heading-XS">Grasa, azúcar y demencia</h3>
<p class="ft-text">Según ha explicado Esteller en la televisión catalana, una de las conclusiones del estudio es que Branyas<span> </span><strong>"tenía dos mutaciones que, pese a ser malas, no le habían provocado ninguna enfermedad"</strong>, lo que explicaría que "tenía mecanismos que la protegían de estas mutaciones". Así,<span> </span><strong>sus valores de grasa y azúcar eran "perfectos"</strong><span> </span>y se mantenía<span> </span><strong>alejada de la demencia</strong>.</p>
<p class="ft-text">En concreto, la analítica incluyó su genoma, pero también análisis transcriptómico, metabolómico, proteómico, microbiómico y epigenómico de diferentes tejidos.</p>
<p class="ft-text">Los investigadores identificaron<span> </span><strong>variantes genéticas raras<span> </span></strong>a través de la secuenciación completa del genoma, y constataron su excepcional perfil lipídico en sangre, con<span> </span><strong>niveles de colesterol bueno (HDL) muy elevados y de colesterol malo (LDL) muy bajos</strong>.</p>
<p class="ft-text">Por su parte, el análisis de las heces ha corrido a cargo de los científicos del<span> </span><strong>Instituto de Investigación Biomédica de Girona<span> </span></strong>(IDIBGI), dirigidos por el doctor<span> </span><strong>Xavier Aldeguer,</strong><span> </span>quien considera que<span> </span><strong>la microbiota se comporta como un órgano<span> </span></strong>al tener capacidad de metabolizar proteínas o grasas. Pues bien. El estudio de Branyas ha desvelado que la mujer contaba con un número de<span> </span><strong>bifidobacterias</strong>, que son antiinflamatorias, correspondientes a la<span> </span><strong>edad infantil<span> </span></strong>y que, según han explicado en TV3, "podrían estar ligadas a la<strong><span> </span>ingesta de lácteo</strong>s", ha añadido.</p>
<p class="ft-text">Maria Branyas no tenía exceso de<span> </span><strong>azúcar en sangre</strong>, lo que evitaba el riesgo de diabetes u obesidad, y observaron que sus genes mantenían<strong><span> </span>bajo control las infecciones y la regulación autoinmune</strong>, además de presentar bajas concentraciones de glicoproteínas A y B, lo que indica un<span> </span><strong>perfil inflamatorio saludable</strong><span> </span>que evita la presencia de enfermedades inflamatorias sistémicas.</p>
<p class="ft-text">Esto ayuda a comprender su<span> </span><strong>buen estado de salud</strong>, ya que al final de su vida tan solo sufrió<span> </span><strong>sordera<span> </span></strong>y<span> </span><strong>dolor articular</strong>, mientras que su lucidez se mantuvo hasta casi al final, con un bajón que tuvo lugar solo en los últimos meses.</p>
<h3 class="ft-helper-spacer-t-xs ft-helper-fontSize-heading-XS">Un estilo de vida saludable</h3>
<p class="ft-text">Los investigadores comprobaron que Maria Branyas seguía una<span> </span><strong>dieta mediterránea</strong><span> </span>que incluía la ingesta de<span> </span><strong>tres yogures al día</strong>, y están convencidos de que esto contribuyó a mantener sanas sus bacterias intestinales.</p>
<p class="ft-text">"Le gustaba<span> </span><strong>andar</strong>,<span> </span><strong>no bebía</strong><span> </span>alcohol ni<span> </span><strong>fumaba<span> </span></strong>y estaba<span> </span><strong>acompañada<span> </span></strong>muy a menudo por su familia, con lo cual no se sentía aislada y esto evitaba la demencia", ha explicado Manel Esteller.</p>
<p><span>Según los autores de este estudio, los resultados servirán para tratar de elaborar "tipos de patrones prebióticos asociados a la longevidad", y además, trabajarán para conseguir </span><strong>"fármacos útiles en la lucha contra el envejecimiento</strong><span>, junto a la definición de dietas adecuadas".</span></p>
<p><span>El estudio refuerza, asimismo la idea de que el </span><strong>envejecimiento y la enfermedad</strong><span> pueden, bajo determinadas condiciones, </span><strong>desvincularse</strong><span>, lo que "desafía la percepción general de que están inexorablemente ligados", explican.</span></p>]]></content:encoded>
